Modeling Chemical Reactivity in Ionic Detergent Micelles: a Review of Fundamentals

چکیده

Ionic detergent micelles have the capacity to solubilize organic substrates, interact selectively with counterions, repel coions, exhibit partial “dissociation” of the counterions, grow in size with added salt, affect the position of chemical equilibria, accelerate or inhibit the rates of chemical reactions, modulate photochemical reactivity and determine the dynamics of diffusion or neardiffusion controlled processes. Many of these phenomena can be understood and analyzed quantitatively in terms of relatively simple models for binding, selectivity and electrostatics that often require no knowledge of micellar structure or dynamics (the pseudophase limit), without compromising chemical intuition. An overview is provided of our current understanding of the interplay between micellar structure and electrostatics, selectivity, solubilization, and reactivity and their role in the development of quantitative formalisms for analyzing micellar effects on reactivity and equilibria.

Solubilized proteins Detergent-solubilization of membrane proteins has proved to be a successful approach in many cases. Both ionic and nonionic detergents have been used: the choice of detergent may be crucial for retaining protein activity. Generally, detergents should be used at concentrations above their critical micelle concentration (CMC). Micelles formed above the CMC mimic a membrane en...

Application of isothermal titration calorimetry for detecting lipid membrane solubilization

Isothermal titration calorimetry is presented to be an excellent method of detecting lipid membrane solubilization upon the addition of detergents. As an example, the titration of the nan-ionic detergent C t2EOt, to the lipid POPC is studied. The main contribution to the measured titration heat is the detergent's enthalpy difference upon transfer between micelles and membranes. The detergent ad...

Analyzing the Physico-Chemical Parameters of Detergents and Detergent Mixtures

The most important physico-chemical characteristics describing detergent micelles are the CMC value and the aggregation number. These parameters depend critically on external conditions such as pH, ionic strength, buffer composition or temperature. However, the influence of these conditions on CMC or aggregation number is a priori not to predict, but the most important parameter. This holds esp...
